Lipids are versatile, safe, and scalable, and can address a variety of delivery challenges, but remain underutilized —only 2-4% of marketed oral drugs are designed using lipid formulations. By tapping into this overlooked technology we can impact more than half of all drugs in development.

Technology Overview

Lipid formulations are challenging to design due to unreliable correlations between in vitro tools and in vivo performance. Pharmacokinetic results often vary significantly between animal models and humans, providing limited quantitative insight into selecting lipid excipients and their optimal ratios. As a result, formulation decisions rely heavily on trial and error, with lipid formulations often discovered serendipitously rather than through a rational design approach.​

At Tret Therapautics, we have developed a lipid formulation technology designed to overcome these limitations. Our algorithm systematically predicts lipid degradation in the gut based on lipid type, quantity, composition, and species, allowing precise control over lipid degradation rates to maximize drug release and absorption across the intestinal wall. Additionally, the lipid formulation can be optimized for human consumption and validated in animals based on an expected/predicted bioavailability. Our approach removes guesswork by designing lipid formulations to achieve targeted pharmacokinetic outcomes from the start, streamlining development and conserving time and resources. 

 

By leveraging the inherent advantages of lipids, our platform has the potential to shift the paradigm in formulation development, making lipids the first choice rather than last, taking full advantage of their safety, affordability, and broad applicability across various drug classes.
